观察研究急性饮酒前后,小鼠海马区锋电位(Spike)的放电频率及峰峰间期等的变化,描述并分析清醒状态及急性饮酒后小鼠海马区神经元电信号的特征差异,评价急性饮酒对小鼠海马区记忆功能的影响。以ICR小鼠为实验对象,分成急性饮酒组(P组)和生理盐水对照组(C组)。在小鼠海马CA1区植入8通道微电极阵列,利用神经信号处理采集系统Cerebus记录急性饮酒前后小鼠海马CA1区神经电信号,分析比较海马区神经元放电频率以及锋电位间隔直方图(Interspike Interval Histograms ,ISI)的变化。P组与C组比较,P组平均放电率小于C组;与清醒状态下比较,急性饮酒后小鼠海马区神经元放电频率变低,然后随着时间逐渐增强,慢慢恢复;ISI值从较为集中变为较为分散。急性饮酒后小鼠海马区神经元自发放电在放电频率、ISI等神经信号的特征方面存在明显差异。急性饮酒可抑制小鼠海马区神经元放电,抑制小鼠记忆功能。
Abstract:
To study the characteristics and evaluate the effects of acute alcohol consumption on memory function, weobserved the discharge frequency and interspike interval (ISI) changes of spike potentials from the hippocampal region of the mice.ICR mice were divided into 2 groups (n=8), including acute alcohol consumption group (P group) and saline control group (C group).The 8-channel microelectrode arrays were implanted in the CA1 region of the hippocampus of mice and then the signals before and after acute alcohol drinking were recorded by Cerebus, a signal processing system.At last, the changes of discharge frequency and Interval Histograms Interspike (ISI) of the hippocampal neurons were investigated.The mean values of firing rate were lower in control group than those in alcohol consumption group. In the alcohol consumption group, firing rate began to decrease right after the injection. After 20 min, the firing rate began to increase gradually. With the time from pre-injection to 80 min post injection, the ISI values became more dispersed. There are obvious differences in the characteristics of the discharge frequency and ISI in the hippocampus of mice. Acute alcohol consumption can inhibit the neuron discharge in hippocampus and the memory function of mice.
