摘要:
为探究利用显微共聚焦拉曼光谱仪辨别经瑞氏-姬姆萨染色后的肿瘤细胞与正常细胞的可行性。应用拉曼光谱技术在532 nm波长激光下分别检测经瑞氏-姬姆萨染色技术染色的肿瘤细胞与正常细胞,对比6种肿瘤细胞、正常细胞和瑞氏-姬姆萨染料的拉曼光谱特征峰值,正常细胞和肿瘤细胞在208~215 cm-1、278~285 cm-1、336~341 cm-1峰位均有拉曼特征峰。发现6种肿瘤细胞的拉曼光谱峰值和瑞氏-姬姆萨染料的拉曼信号峰值均相同。经分析认为,检测到的均为各种染料的拉曼信号而非6种肿瘤细胞的拉曼信号。因此,无法用显微共聚焦拉曼光谱仪区分经瑞氏-姬姆萨染色后的肿瘤细胞与正常细胞。
Abstract:
To explore the feasibility of using micro confocal Raman spectrometer to discriminate tumor cells and normal cells stained by Wright-Giemsa dye,we used Raman spectroscopy to detect the tumor cells and normal cells with Wright-Giemsa stain under 532 nm wavelength laser, and compared the Raman spectral characteristic peaks of 6 kinds of tumor cells, normal cells and Wright-Giemsa dye.We found that the Raman peaks of 6 kinds tumor cells were the same as those of Wright-Giemsa dye. The results showed that the detected Raman signals were the Ramam signals of Wright-Giemsa dye, not those of 6 kinds tumor cells. Therefore, it is impossible to discriminate tumor cells and normal cells by using micro confocal Raman spectrometer.
