摘要:
磁动光学相干弹性成像(magnetomotive optical coherence elastography,MM-OCE)可通过检测负载磁性粒子的组织的微小振动,分析振动信息,提取能反映组织生物力学特性的参数。本研究利用螺线管磁场激励与光学相干层析成像系统采集模块搭建了MM-OCE研究平台,通过跟踪负载磁纳米颗粒的组织在扫频信号激励下的振动响应,确定组织的机械共振频率,从而半定量地表征组织的生物力学特性。研究结果显示,当自制仿体中明胶含量增加时,测得的共振频率增大,反映其硬度增大;对离体角膜进行戊二醛胶原交联硬化处理,随着戊二醛浓度提高,处理后的角膜的共振频率也相应提高,表明共振频率可反映角膜硬度的差异。上述结果表明,该研究可评估角膜组织的力学特性差异,在临床中具有较大的应用潜力。
Abstract:
The magnetomotive optical coherence elastography (MM-OCE)can detect the tiny vibrations of the tissue loaded with magnetic particles, analyze the vibration information, and extract the parameters that can reflect the biomechanical properties of the tissue. We used the solenoid magnetic excitation and optical coherence tomography acquisition to establish the MM-OCE research platform, which records the vibration response of the tissue loaded with magnetic nanoparticles under the excitation of the sweep signal to determine the mechanical resonance frequency of the tissue, so as to characterize the biomechanical properties of the tissue semi-quantitatively. The results showed that the resonant frequency increased when the portion of gelatin was enhanced in the gelatin phantom, indicating the increase in stiffness for the gelatin phantoms. After cross-linking treatment with glutaraldehyde collagen on the ex vivo cornea, the resonance frequency of ex vivo corneas increased with the increase of glutaraldehyde concentration, indicating that the resonance frequency could reflect the difference of corneal hardness. This research can evaluate the difference of mechanical properties of tissues and has great potential for clinical application.
